SERPINE2 feedback regulates EGF/EGFR signaling in human papillary thyroid carcinoma cells

Thyroid cancer (TC) is the most prevalent malignant tumor in endocrine system. Serpin peptidase inhibitor clade E member 2 (SERPINE2) closely associated with metastasis. The aim of present study was to investigate whether SERPINE2 forms a feedback loop epidermal growth factor (EGF)/EGF receptor (EGFR) that regulates cellular processes human papillary thyroid carcinoma (TPC‑1) cells. Reverse transcription‑quantitative PCR and western blotting were utilized analyze expression SERPINE2. Cell proliferation ability detected cell cytotoxicity assay kit (MTT) by clone formation assay. markers, including proliferating nuclear antigen Ki‑67, also investigated proliferative activity TPC‑1 Besides, migration invasion analyzed wound healing Transwell assays, respectively, while apoptosis TUNEL staining. results showed increased TPC cells, EGF/EGFR regulated each other. Furthermore, overexpression silencing proliferation, migration, apoptosis. an EGFR blocked effects on aforementioned biological processes. Therefore, confirmed formed positive regulate possibly providing novel insights into potential therapeutic targets TC.
